淫羊藿苷配伍人参皂苷Rg1对骨髓间充质干细胞成骨分化的作用Effects of Icariin Combined with Ginsenoside Rg1 on Osteogenic Differentiation of Bone Marrow Mesenchymal Stem Cells

摘要(Abstract):
研究淫羊藿苷和人参皂苷Rg1不同浓度配伍对骨髓间充质干细胞(BMMSCs)成骨分化的作用。采用Cell Counting Kit-8(CCK-8试剂盒)观察淫羊藿苷和人参皂苷Rg1单独用药及不同浓度配伍对C57小鼠骨髓间充质干细胞增殖情况的影响;在成骨诱导第16、30 d后用茜素红进行染色,观察成骨细胞矿化能力;采用反转录实时定量PCR(RT-PCR)及蛋白质印记(WB)检测成骨细胞标记性蛋白人骨形态发生蛋白BMP2、RUNX2、OPN、OCN的表达情况。淫羊藿苷和人参皂苷Rg1单独用药最佳浓度分别为1μmol/L和10μmol/L;两者配伍最佳浓度分别为1μmol/L和20μmol/L。与单独用药相比,配伍能够明显促进诱导细胞的钙化作用并且上调成骨细胞标记性蛋白BMP2、OCN、OPN、RUNX2等的表达,淫羊藿苷和人参皂苷Rg1配伍能够促进骨髓间充质干细胞成骨分化。
